Unable to read raw pixel data for a single pixel

AcquireOnePixel() is a MagickCore method. You can access it with MagickLib::AcquireOnePixel(). For Magick++, use Magick::Pixels::getConst() to acquire pixels.

For the command line use

   convert image.png  -crop 1x1+3+5 txt:-
to get the 3,5 pixel
